TITLE : Fluorimetric investigation of secondary liuninescenen of
lymphocytes from irradiated rat lympla nodes
SOURCE: Radiobiologiyat v. 5, no. 5, 1965, 685-689
TOPIC TAGS: experiment animal* irradiation effect,, aall pIrlysiology,
luminescence
ABSTRACT-: Albino male rats weighing 200 g were X-irrftdiated (RUP-200-
20-5 unit 170 kv, 15 ma, 0 cm focal length, 0.8 cm Al + O.K cm Cu
filters, 44 r/min) with a NO r dose and killed 5 to 10 min.'2, 3, and
12 hrs later. Lymph nodes taken from the animal's groin were cut open
and lymph tissue was carefully removed and suspended In a physiologioal
solution. The tissues were stained with acridine oriingo to determine
secondary luminescence of average and small lymphocyte nuolei with a
microscope-eytofluorimeterp sensitive to the green I)Llad of the spectrum
Luminescence was excited by a light wave length of 06 millimicrons and:
iwas measured in the 510 to 550 millimicron range. Arithmotical mean

values for nuclear luminescent intensity were computed for cells in
experimental and control samples. Results show that with a microfluor-
imeter small differences in fluorescent inten3ity can be detected in
the lymph nodes of normal animals which appar-antly reflect the aging
processes and cell degeneration, In 5 to 10 min following a 500 r dose
a statistically reliable increase in intensity of lymVqocj-t(b nuclei
luminescence is found. Two hours latex, a considerable mmbor of cells
appear with bright orange luminescence of the nuclei reflecting the
pyknotic process and also intenflification of the relgitive i;:,radiation
effect which reaches a maximum after 3 hrs. The level of the relati7e
irradiation effect after 12 hrs is reduced to the level obsorved after
2 hrs; a possible explanation for this may be that the damaged cells are
destroyed by macrophages and lytin agents of the orgi)n. The authors
t1express their gratitude to M. IT. Meysel.. Corresponding Member of AN
SSSR, for his guidance and helpful advice, V, Yu. Urbakh for consulta-
tions on statistical processing of data, and A. A, Zotikov for his
assistance." Orig. art. has: 1 table and I figure,
