MEMORANDUM FOR: THE RECORD

The purpose of:this subproject 47 is to provide funds to continue
a research program 'being carried out by Carl C. Pfeiffer, MD.
2. The nature of the investigations to be carried out under the
direction of Dr. Pfeiffer enc mpasses pharmacological and clinical testing
in animals and human volunteers of chemicals and biochemicals which alter
,behavior. Certain phases of other problems of interest to TSD will be
investigated.
3. Dr. Pfeiffer is located at the Nevt Jersey Neuro-Psychiatric
Institute in the Bureau of Research in Neurology and Psychiatry, Princeton,
New Jersey.
4. The funds for this project will be transferred using the

5. The cost of this program for a period of one year beginning 1 June
1962 will not exceed $25,000.00. To this SWII must be added $1,0.00.00
representing a 4% service charge to th

INTRODUCTION:
Our research efforts have received local attention and approbation
during the past year so that we now have excellent rapport with the staffs
of both the Institute and theB6rdentown-Reformatory. We now have 10
volunteers per week from the latter and we have a Clinical Investigative
Unit which will house 80 schizophr lies (present census 45 patients).
Current Status of the Pharmacology Unit
The laboratory is still undergoing minor construction, but material
progress has been made in the past year. The section for animal screening
and conditioning is complete. The sections for animal neuropharmacology
and clinical pharmacology have been constructed and put into operation,
andan electronic workshop has been set up. All these units are actively
at work in human and animal experiments as outlined below. The T.,.nilities

of the clinical unit---experimental ward---are nearing completiOnl and
are already in partial use. Still in varying stages of construction are
the biochemical pharmacology and medicinal chemistry units.
Personnel: Sin de the last report, the section has been joined by Drs.

AileZrand as planned. It is anticipated that two to three
more investigators at the doctoral level will be needed, probably one
more clinical pharmacologist, a medicinal chemist, and a biochemical
pharmacologist. Two engineers have been employed for design and con-
struction of new equipment and six lab assistants are now fully trained
to their various jobs.
Human subjects: Liaison with:NaZe'rseifState Reformatory -,Bordentown
has been excellent, and a full complement of volunteers from that insti-
tution has been freely available. This group may be a better group for
this kind of research, since they may be closer to the normal population
than were the Atlanta prisoners, the latter being recidivists and older
than the BOrdentoWn'group. In addition other human volunteers have
been recruited, including female subjects, so that the population of
"normal" subjects available is now much more nearly representative of
the population at large. In addition, it has been possible already to
begin explorations with the psychotic patients from the experimental
ward, and some interesting results are already forthcoming (see below).
Equipment: 'Since the last report, the section has obtained a stereo-
taxic apparatus, a Grass Model III EEG and a Sanborn 2000 magnetic
tape Data Recorder.. In addition, we have constructed two new integrators
for quantitative EEG; these are modeled after the DioNocki Integrdtoe;
but due to some circuit changes are more linear', and have the advantage
of using all Ur_A-Alade parts, so that maintenance is simplified. Two
more integrators are under construction which will albw simultaneous

-integration of four leads, one from each lobe of a hemicranium. Also
under construction is a quantitative spectral analyzer for use with the
tape recorder in analyzing scalp EEG of humans as well as electrical
measurements from implanted electrodes in cortex and deeper structOres
in brains of animals. Data will be entered on punch cards and processed
by a digital computer. Target date for beginning calibration and stan-
dardization is June 1, 1962. These new approaches allow determinat5on
with great accuracy of effects of various classes of compounds and the
interactions of such agents.
A Barber Colman gas chromatograph has been installed and a technician
is being trained to operate it. Thir makes possible correlative studies
of changes in brain constituents, changes in behavioral state, and electro-
physiological data. This method is far more sensitive than previous
techniques, and allows accurate measurements to be made from much smaller
brain fractions. A Beckman pH meter-and microburette have also been
obtained for study of cholinesterase activity, since it appears that
psychotomimetic effects frequently if not always involve atropine-like
drug action (Pfeiffer, C.C., et al. Neurology 9:24925O, 1959)
Recent and current projects: These new techniques, together with the
unique availability of human subjects both normal and abnormal, make
possible a wide range of investigations on any class of compounds with
action on the central nervous system. These can begin at the Most basic
level and move progressively to a level of sophistication beyond that of
ali7.cst any other setting. Sore idea of the directions currently being
talen are indicated in recent and current work. Enclosed a e-reprints of
publications during the past year. The following are also accepted for
publication and will appear in the next few months:

the following are under_way: A number of tryptamine derivatives are
being explored for psyehotomimetic and other effects; 4-hydroxy-a-methyl
tryptamine, 4-methyl-a-methyl tryptamine, 64ydroxy-N,N-dimethyltryptamine
(The di-methyl analogue of Szara's compound.),. prolyltryptamine, psylocin,
and acetyl tryptamine. Results will be reported as they accrue. The
central actions of,y-aminobutyrie acid are being indirectly explored by
investigati;g y-butyrolactone and y-hydroxybutyrie acid. Unlike GABA,
these compounds readily cross the blood-brain barrier, and the:latter is
a potent hypnotic agent and about as active and positive as is chloral hydrate.

� Screening of tryptamiqe compounds in animals and man for hallucino-
genic action.
� Completion of the study of the use of riboflavin as a trace chemical
to prove the ingestion of medication in a clinical setting.
3. Further studies on the use of phenoxybenzamine and other adrenergic
blocking compounds as antidotes to-LSD-25.
4. Further refinement of the use of quantitative brain wave studies to
diagnose schizophrenia and to determine the action of hallucinogens.
5. Selection of one or more of a series of stimulant diamines for chronic
toxicity studies in animals so that they can be tested in normal human
volunteers for possible stimulant action.
6. A search in brain extracts of rabbits treated with Tofranil (imipra-
mine) to determine if any stimulant amine accumulates as a result of
degradation of the drug molecule in the body.
7. Determination of the anti-cholinesterase and aminoxidase inhibitory
effect of the stimulant diamines.
8. Assay oface0:holine content of various portions of the biain after
prolonged administration of deanol.
9. The assay in man of an intravenous analgesic solution composed of
morphine and choline or morphine and carnitine wherein the. pharma-
cologically neutral quaternary compound prevents the engagement of

It Is proposed to conduct screening and range finding tests on volunteer
with chemical compounds of known behavioral activity. Studies will be
conducted on derivatives of selected biologically active compounds to deter
mine their action on the central nervous system of if-an.
In addition to these investigations, the following work will be undertaken:
� 1. Pharmacological testing will be conducted on animals
utilizing candidate hallucinogenic chth-nicals.
2. Studies will be conducted on compounds of relativly
simple configuration to assess their ability to inhibit trained

volunteers for possible behavioral changes utilizing compounds

1. 'The purpose of this subproject 47 is to provide funds to continue
a research program being carried out by Carl C. Pfeiffer, MD.

It is proposed to conduct screening and range finding tests on volunteers
with chemical compounds of known behavioral activity. Studies will be
conducted on derivatives of selected biologically active compounds to deter-
mine their action on the central nervous system of man.
In addition to these investigations, the following work will be undertaken:
1. Pharmacological testing will be conducted on animals
utilizing candidate hallucinogenic chemicals.
2. Studies will be conducted on compounds of relativly
simple configuration to assess their ability to inhibit trained
behavior in animals utilizing the condition avoidance response
3. Studies will be conducted of compounds in normal human
volunteers for possible behavioral changes utilizing compounds
provided by the sponsor.

of the ompOunds at the Atlanta federal Penitentiary under the direction of

this time the human testing will be organized and progressing in New Jersey under

team in the Nelot Jersey location.

Proposals A, 8, and C are a continuation of th ork now in progress which
is familiar to the officers of TheSiVSaVai% . from our annual and s'emi-
annual reports. Under C thrt:,.source of human volunteers will be the 100 reformatory
inmates (ages 18 to 28) who .?re stationed on the grounds of the Institute. If 16
volunteers, aged 21 to 28; asre not obtained frail this group for a weekly session
of drug testing, we will have one session of eight subjects at the institute and
one session at the New jersey-State Prison in Trenton. %Commissioner erainberg has
given his permission.
Under D we now have perfected and plan to use at the New Jersey Laboratory a
quantitative integrator for drug studies in both man and animals.
Adult unanestheti2ed, trained animals are used. Under anesthesia small gold-
plated wood screws are used as dural electrodes. These are lightly engaged in the
calvarium and held firmly in place with methacrylate with the permanent lead
attached. The recording leads are bipolar from the left and right frontoparietal
lobes of the brain. The animals are in a sound attenuating box and sit quietly for
the entire 1 to 2 hour session after a few training periods.---
A Orohocki integrator is attached to one lead. This device rectifies the brain
waves and by a chopping process, counts the area under the rectified curve. A
signal is made at a pre-set degree of sensitivity. The number of signals per unit
time is a measure of the degree of-electrical activity or "electroeenesis." The
number of pulses or signals is directly correlatable with the area under the curve.
The constancy of the electrogenesis with elapsed time can be calculated and treated
by the usual statistical methods. In this type of analysis, periods as long as
five minutes can be used as a baseline or control ad thus transient artefacts are
eliminated. The action of drugs is usually measured at 1-5 minute intervals after
intravenous injection and the drug action is expressed as change in percentage
electrogenesis from the control preinjection period. AroLsal or stimulant drug
effects are recorded as a decrease while sleep (sine waves) produce an increase in
electrogenesis.
E. Annual conference on neurobiology of psychoses. Most previous conferences
of this type have been poorly planned, and the group has been too large and unwieldy.

The participants will be limited to 12 or l5. The record will be in the form
of the individual's own notes and the notes of a scientifically trained secretary.
The summarizing secretary will change with each three-hour session. In a two-day

etiological factors listed would determine the agenda and the priority of the factors

If the grant can be activated as of June 1, 1950, we shall be able
to order certain items of equipment and thus decrease the delay which .
nay result in changing our laboratories. I shall begin work in New. -
Jersey as of July 1, 19S0 if you or the officers of the
tV..42* have any questions, I shall be glad to appear before your group
to explain or justify any of the budgetary items or expand on the
details of the proposed research.

MEMORANDUM FOR: THE RECORD
: MKULTRA, Subproject 4?
1. TI)eurpose of this subproject is to provide funds for
the continuation of the program being carried out at Emory Unive
sity under Carl CPfeiffer, M.D. The scope of this work basically
covers the development and testing of new chemical compounds which
are hallucinogenic or otherwise affect the central nervous system of
humans. Certain phases a other problen-is, of interest to TSS/CD will
be investigated.
Z. The nature of the investigations to be carried out under the
direction of Dr.1 Pfeiffer encompasses the synthesis pharmacology
and pertinent biochemistry of potential hallacinogenic chemical corn-
pounds. Testing and evaluation will be conducted in normal human
volunteers.
3. The funds for this project will be ttansferr d using the

4. The cost of this program for a period of one year beginning
1 June 1959 will not exceed $4'1%378.00. To this sum must be added

1. Continued testing program in animals and man of the potential hallucinatory chemi-
cals which have been synthesized by or which become available from
commercial laboratories. ow has 30 chemicals awaiting testing in man.
2. 2-dimethylaminoethanol in preliminary trials modifies the ISO-25 model psychosis
in that the onset of symptoms is delayed but in the individual subjects the visual
hallucinations are more frequent. A double blind test of rigid design will be
set up to quantify this effect.
The possible occurrence of 2-dimethylaminoethanol in natural foods will be deter-
mined by steam distillation and paper chromatography.
I. Tests will be made on tamitine and its tertiary amine derivative to ascertain its
possible role as a neurohumor in the central nervous system.
5. The D and 1. forms of lactylcholine have been synthesized and will be extensively
tested in animals to ascertain the possible similarity to acetylcholine. Other
natural occurring alpha hydroxy acids will be esterified to choline to determine
their potency.
6. Continued testing in man and animals of those compounds which may be submitted

the continuation of th-e-oograrn being carried out at Emory Uni-
versity under Dr. Carl (C. Pfeiffer. The scope of this work is

1.t.The purpose o! this project is to provide funds to
coatinue the research program conducted by Dr. Carl Pfeiffer

the knowledge and facilities available to its director in pro-
viding answers to specific questions of direct interest to the
Agency. In addition, those areas of basic knowledge required
to provide these practical answers swill be-developedj It

A second phase will be represented by
an extensive effort on behalf of Dr.
Pfeiffer to find a solution to the re-
quirement for an anti-interrogation
drug. This requirement was laid on
TSSiChemical Division by another
�si
Agency component. �
A third phase of the program will
provide screening and evaluation
of compounds submitted by TSS/CD,
including the phases involving tests
\�in human volunteers.
The funds for this project will be transferred using

cutout in in the usual manner.
4. Costs for this program for a period of one year
beginningl June 1956 are estimated to be $47,403.00, to
which must be added a four percent (4.-rc) fee to the

annual basis of 211 monies received from the Fund. Also,
it has been requested that any unexpended funds be returned

which has been conducted during the last two years by Dr. Carl
C. Pfeiffer under thelinominal title, "The Plwrmacological Screening
and Evaluation of Chemical Compounds Having Central Nervous
System Activities."
The program at Emory is necessarily divided into several
parts, some of which are supported by grants other than that of
